چکیده

Purpose: To study the effect of dexmedetomidine (Dex) on AKTERK signaling pathway and EMT- related proteins in high glucose-induced apoptosis human kidney tubular epithelial cells.
Methods: HK-2 cells were assigned to control, high-glucose Dex groups. Levels ROS determined using live cell station. Flow cytometry was used measure cycle while Western blot applied assay levels PI3K, Akt, p-Akt, ERK p-ERK.
Results: The concentrations markedly reduced group, relative glucose group (p < 0.05). Apoptosis P13K protein significantly lower groups than their corresponding control levels. In glucose-treated cells, AKT expression downregulated, p-AKT Protein expressions p-ERK values, but accentuated 0.05).
Conclusion: mitigates increases proportion G1 phase, reduces EMT via a mechanism regulation pathway-associated proteins. provide insights into development drugs for treatment diabetic nephropathy.

Effect of zinc on high glucose-induced epithelial-to-mesenchymal transition in renal tubular epithelial cells.
Zinc (Zn) as an essential dietary element has been indicated in a number of protein functions in the prevention of numerous types of epithelial-to-mesenchymal transition (EMT)-driven fibrosis in vivo. However, relatively little is known regarding its effect in the EMT of the renal tubular epithelial cells, which play an important role in renal tubulointerstitial fibrosis and is an important com...
متن کاملNicotine-Induced Apoptosis in Human Renal Proximal Tubular Epithelial Cells
BACKGROUND Nicotine is, to a large extent, responsible for smoking-mediated renal dysfunction. This study investigated nicotine's effects on renal tubular epithelial cell apoptosis in vitro and it explored the mechanisms underlying its effects. METHODS Human proximal tubular epithelial (HK-2) cells were treated with nicotine. Cell viability was examined by using the WST-1 assay. Intracellular...

BACKGROUND/AIM To investigate the role of kidney injury molecular 1 (KIM-1) in high glucose-induced autophagy and apoptosis in renal tubular epithelial cells. METHODS Human renal tubular epithelial cells (HK2) were treated with normal glucose (NG, D -glucose 5.6 mmol/L), high glucose (HG, 30 mmol/L), high osmotic (HO, D-glucose 5.6 mmol/L + D-mannitol 24.4 mmol/L), HG + KIM-1 siRNA, HG + siRN...
متن کاملOxidative Stress-Activated NHE1 Is Involved in High Glucose-Induced Apoptosis in Renal Tubular Epithelial Cells
PURPOSE Diabetic nephropathy (DN) is a prevalent chronic microvascular complication of diabetes mellitus involving disturbances in electrolytes and the acid-base balance caused by a disorder of glucose metabolism. NHE1 is a Na⁺/H⁺ exchanger responsible for keeping intracellular pH (pHi) balance and cell growth. Our study aimed to investigate roles of NHE1 in high glucose (HG)-induced apoptosis ...
